氨基柱高效液相色谱法检测大豆提取物中的磷脂酰胆碱

摘要:

建立一种利用高效液相色谱检测大豆提取物中磷脂酰胆碱的简便方法。色谱柱为氨基色谱柱(250 mm×4.6 mm,5 μm),流动相洗脱条件为20%(V/V)乙腈水溶液等度洗脱,检测波长为206 nm,流量为0.8 mL/min,以峰面积为指标进行定量分析。结果表明,磷脂酰胆碱的保留时间为5.292 min,峰面积与浓度之间线性关系良好,相关系数(r2)为0.9996;稳定性实验、精密度实验、加标回收率实验表明该方法准确、稳定、可靠。按照此方法检测得到,经无水乙醇提取的大豆提取物中磷脂酰胆碱的含量为16.31%。该方法简单、快速、准确,可丰富卵磷脂中磷脂酰胆碱的检测方法,为其高效检测提供参考。


Determination of phosphatidylcholine in soybean extract by high performance liquid chromatography with amino column

Abstract: The method for determination of phosphatidylcholine in soybean extract by using high performance liquid chromatography (HPLC) equipped with an amino column (250 mm×4.6 mm, 5 μm)was established. 20% acetonitrile aqueous solution (V/V) was used as mobile phase for isocratic elution. The detection wavelength was 206 nm and the flow rate was 0.8 mL/min. The peak area was used for quantitative analysis. The results showed that the retention time of phosphatidylcholine was 5.292 min and the peak area had a good linear relation with the concentration for the correlation coefficient was 0.999 6. The accuracy, stability and reliability of this method were certified through the precision test, stability test and standard recovery test. According to this method, the content of phosphatidylcholine in soybean extract extracted by anhydrous ethanol was 16.31%. The method established is simple, fast, accurate, which is expected to enrich the detection methods of phosphatidylcholine in lecithin and provide reference for the efficient detection of phosphatidylcholine.
