RP-HPLC法测定泰妙菌素与多西环素的含量

摘要:

概述了RP-HPLC法测定复方泰妙菌素可溶性粉中泰妙菌素和多西环素含量的方法。采用Shim-packCLC-ODS柱(150 mm6 mm,5m),流动相为80%乙腈-0.7%磷酸氢二铵(体积比为0.3:0.7),并用氨试液调节pH至8.00.1。流速为1.0 mLmin-1,泰妙菌素检测波长为208 nm,多西环素检测波长为272 nm,柱温为40℃。在此色谱条件下两者能完全分离,泰妙菌素和多西环素在10gmL-1~100gmL-1的范围内浓度与峰面积呈良好的线性关系,回归方程和相关系数分别为:A=47505.7+1003.32,R=0.9999;A=47357+5587.3,R=0.9999。泰妙菌素和多西环素的回收率分别为99.51%~99.84%、99.75%~100.2%;RSD分别为0.30%~0.75%、0.45%~0.80%。实验表明,该方法简便、准确,可用于复方泰妙菌素可溶性粉的含量测定。


Determination of tiamulin and doxycycline in compound tiamulin powder by RP-HPLC

Abstract: The determination method of tiamulin and doxycycline in compound tiamulin soluble powder by RPHPLC was introduced. In this method, Shim-pack CLC-ODS column (150 mm×6 mm, 5 μm) was used, the mixture of 80% acetiontrile-0.7%(v/v) dihydrogen ammonium phosphate (0.3:0.7) was used as the mobile and it was adjusted to pH 8.0±0.1 by ammonia liquor, the flow rate was 1.0 mL/min, the temperature of the column was 40℃, and the detection wavelength of tiamulin and doxycycline hydrochloride was 208 nm and 272 nm respectively. Under the adopted chromatographic condition, the tiamulin and doxycycline could be completely separated. The standard curves for tiamulin and doxycycline were linear in the concentration range of 10 μg/mL~100 μg/mL and their regression equations were A=47 505.7ρ+1 003.32 and A=47 357ρ+5 587.3 with their corresponding correlation coeffcients of 0.999 9 and 0.999 9. The recoveries of tiamulin and doxycycline were 99.51%~99.84%, 99.75%~100.2% with RSD 0.30%~0.75%, 0.45%~0.80% respectively. The experimental results show this method is convenient, rapid, accurate, and it can be used to control the quality of compound tiamulin soluble powder.
